[00:00:05] >> Well welcome everybody to another edition of The Fall 2020 minute fans webinars series My name is David Godfried and I want to welcome you nor my our normal host and mc Paul Joseph is on leave this week so I'll be hosting and handling the mc duties so as you may know no fans are sponsored by the by seen it which is the Southeastern nanotechnology infrastructure corridor or which itself is a partnership between Georgia Tech's Institute for electronics and its acknowledging in the joint School of Medicine of science and engineering in Greensboro North Carolina. [00:00:43] You can find out more about scenic at our website which is listed there on the left left hand side so it's a real pleasure to have a speaker with us a Professor Jones and away from j.s. center and particularly the University of North Carolina Greensborough Professor Waite is both bachelors and masters degrees and Applied Chemistry at East China University of Science and Technology in Shanghai before coming to the United States to pursue his Ph d. degree in chemistry at the University of Pittsburgh after completing his doctorate in the state at Pittsburgh for a post-doctoral fellow ship before joining the c.f. d. Research Corporation where he worked for a number of years and then moved to the joint school in a science and engineering in 2013. [00:01:33] And I will turn it over to him for his talk on integrated nano plasma monic biosensors or protein biomarkers and I will stop sharing all that use here Ok thank you for your introduction and also I'm very many you know have all been Trevithick opportunity to share our research at 13000 and. [00:02:01] Yeah you can see my title for these park it's an integrated and normal Prada monic biosensor for protein biomarker. First you know about Jason and. Yeah I'm in the National Science Department belongs to. C.g. but I actually Jason there is. A joint program between 2 universities you can see g. and the and c. and t. So it's built on the strength of 2 university in the basic science and the union here in we hope actually to offer some you know rate of across this program so we only have. [00:02:44] Students Currently we have about 120 students halfhour. Between departments and at Hotel 16 fact m.m.s. also submitted to 2 departments. It's the building I think it's the building itself a spends about $65000000.00 and the sum of facilities as well. So Ok let me just get into my outline of these talk 1st I work a little bit about introduction diagnosis and the problematic sense in. [00:03:23] How people would be valid problematic since and with like a part of the virus background some of the. Science about save the problem rather than stand the localizer it's part of my residence then I will mostly just introduce what a project we are doing try to integrate Prada Mannix. [00:03:48] Sensing and use in the same device actually also facility it. And then it's delivery to the sensing area so we are talking about how we design. A sensor and the use in some a scene in a simulation more the new tool and the fabrication a device assuring some of the results for the device correct translation and that I'm on streatch protein capping and it actually comparable protein markers 1st of all we know that's a diagnosis it's very important if it can be used for personal and it's a medicine is specially you know right now we are always Persian patient concert essential centered so if a person can just diagnose it by himself or herself then we all large the connection. [00:04:44] Can be used for. For diagnosis and then the doctor basically made to be remotely. Have a treatment to the patient is special rate right now the pandemic actually it's also has been we have seen some of the. Diagnosis devices for the virus based on the. Protein biomarkers as well so the part about the virus I think yeah big dreams made over small stuff nowadays for example here are people already tried to create a product group dark which actually grew cause power sensor integrated with a smartphone then 8 already on the market then with a drop of blood you may be able to just you need into the morning to the sugar level or implant and this is related to a lot of disease such as that is and then on the other hand here I'm showing optical sensor a very small pen in prototype prototype. [00:05:59] In this case it integrates like American Aix optics and their sense of shape in a very small promise I it's problematic detector but they in this now they say actually the censorship becomes the key so how we can media tries. Cheap become very important if you want to really get a portable and very small sites detector. [00:06:28] About their safe it's hard to read and we will read in the known very well and so since it's problem rodents basically it's a it's a residence alteration of conduction electrons at the unit of place between an active and the positive or the maturity mature is a stimulated by used in the light which means in this case you know this if it's a problem are we also cautious if it sever the Panama prior written. [00:06:57] Propagate in the x. and the y.. Directions l. on the metal tile actually interface. Normally there to take a lens for their hands $200.00 microns I add to the direction but the decay. But if well this pic and the p.l.o. that it's a showing the enormous feel of particle normally gold or silver particles there will also generates a separate problem resonance but it's a come find a rom the day. [00:07:32] None of us feel safe place in this case we call other areas p.r. there resonance. We weren't there to read them as we wouldn't with their the light and the self is a problem are you actually you actually also lation there that the resonance wavelength of normally related to 2 key factors one is the size or geometric factor over the Nano particle the 2nd of all also the shape the 2nd would be the interface. [00:08:08] Dielectric properties for many we measure as a reflector you next but if for the nominal local anonymous feel on on a particle based localized to the surface of Parliament I didn't nominate the to kill him for he is much shorter in the order $100.00 pins 100 Nonna meter. [00:08:29] Normally they search for key because they are sensing they are 2 complete Grecians their fur their rights one over here it's a very true traditional We called Christian men come figuration which depend rely on their total into the reflection of the light used in their present. This whole ready commercial rights and the Quite a large. [00:08:56] Parking instrumentation for a lot of like surface a biological binding clitic so and the pelham excess study and the sensing of the well there's a 2nd. On the left side people using someone normal fabricated or normal function it's the safest which create low clouds are the safest part of the ordinance and it can be measured as a transmission mode in this case you know there's. [00:09:25] The principle for the for that detection it's a very it's actually the same basically it is the resonance. Light's resonance would be very sensitive. Near the surface dielectric constant changes so here I'm just giving principle over the air s.p.r. sensing you then go the normal sphere Once you have a like at a sub in the layer normally of the biological family or function as a Xing then once there's a lot of biological space of the abandonware there huge increase the thickness of of the biological layer then if you change the reflecting index at the surface then that change over the reflect you next will cause the. [00:10:15] Resonance with limbs shift so in this case you would be about but you will be able to measure the. The bending. Reaction better than your actual Of course depending on the. Concentration of the n. and it's in the solution in this way once you establish the calibration curve curve with a stand then a solution you will be able to care what exactly their concentration in the solution. [00:10:46] You may study based on phenomena that. Was. Reportedly in the $990.00 s. that the surface part of. Normal feature of the cavity in the. Trick with metal field then it accords it would it generates. Lights just mission called extraordinary optical transmission this. Mission actually has because of the resonance between the said the problem so then in this case the optical transmission wavelength also very sensitive through to the few key per meters 1st Still the geometric factor that it's the size of the Nano hole for example a cavity as well as the. [00:11:42] Purity city the 2nd would be the electrical properties of the surface and. The interface so then we would in this case people also develop a different city here it's the non est biosensor in this case their service our problem are you know normally in this case however we called f.n. or resonance for the. [00:12:11] Normal sleep every It's dominates the state of the purity and that I like your property. But for these streets and honestly it normally if you want to get really. Wavelength and wrench in the visible wrench or the the new freedom wrench and therefore the pretty nice sensitivity the Nama speed with is about like a 15 on a meters then there and I skied you were a for example approaching Normally if you nanometers diffusion to disclose more sides gap for 4 sensing or would it be not easy so that's. [00:12:51] Another issue is because of the explosion of the surface gold to the whole biological samples there are huge difference from a by a fluid other like at the tional proteins or human other biological species remained their samples so their idea for was actually we have a goal to try to detect different types of protein by Marx you hope a lot so then we learn and concept use in like are there none of the age and a long there microfluidic channel then we wanted you to invent some normal slits which as. [00:13:35] Capability to not only tacked to any binding reaction this simple Also hopefully can have our summer like a sides. Controlled manipulated. Somebody very only in the protein can get into this into the detection area but are there like bladder sales large particles with a nod or have an interface for the detection so there's a practical chain or G.'s for this design in in the Protein Power sensing 1st how we can design a signature instruction that was strong signal detectable and high signal and the noise ratio and the also the reflecting in x. sensitivity on the other hand you know we would have likely efficiency for the simple and it's delivery the other is of course it's for almost every affinity. [00:14:35] There and we need to use in some a separate function addition and. At least take a read of some of the man specific abandon the device integration of course it's there for long term goal if we really want to get a portable device the component into the connection and the readout but they actually are not yet gather that far so we only talk about. [00:15:02] The signature instruction and that's delivery under some of the detection that. They have fabricated not a problem actually this is. The device concept are the Nano structure basically we have a open nano sleet and there combine away there and there are none of it in the bottom there that gold fear we design it's about Iran like a 200 an animator and then. [00:15:34] Actually it could have been lower the thing is that we design it a 115 on a meter so we have the open about with don't know yet but we have hopefully we can get like a few 100 a nominee to with the new. $59.00 a meters which would have created such a strong monopod the Manics in them honestly and there are that it's like at the quantities and we're not going to talk about this we only log talk about them on alert structure here in this case the hype about the states that are the. [00:16:09] Largest here would just flow over the top of the Micro Channel with a nod to getting into the monolith structure though these say it's the key for their signature instruction how we design this structure Fortunately actually back to few. 50 years ago. Peter man and call it develop a theoretical model to kind of analyze their services problem our generation in the street and honestly it especially in the boundary of that are you actually interface at the Honestly in the cavity so in this case basically the s.p.p. generation results from that stage mechanism 1st it's a purely John Mitchell get diffraction follow the Bible there not show for the 7 the problem our priority. [00:17:02] Polarity Mbali the mode on the flat interface with the airplane wave so that the n.s.a. the based on the plane way of where you stand the light either. At the moment it's the through the Uniphase So then then the strength of the alarm that interface can be calculated can be calculated the use in this model. [00:17:27] But if all Ok is then we then that basic knowledge then we decompose that interface to be 3 interface which contributes to the surface problem or veterans or a well as a p p So the 1st it's an interface with the courts and a go to interface the 2nd once we have any binding or you may be in the air. [00:17:54] Especially the shop. Location it here actually it's their hot spot generates s.p.p. So these 2 basically it's there with the same mechanism by the same and then their boundary of the universe of the chords so we develop the strength of the s.p.p. expression based on the more 2 previous slide soon then we calculated using this s.m. and an item or 2 kind of a to compose and the we calculated the s.p.p.. [00:18:30] Plane way of where you stand light then it. By changing the wavelength light to use in the we went as well as them honestly and the reflector in the index so this stuff basically it's at it's there are reflected in the x. and the actual or minus a little weight and then the wavelength used in the light so we calculate that the s.p.p. generation efficiency alarm. [00:18:58] Wavelength and. Operates at the perimeter so we found actually once we find that reflecting index of any binding in the surface it's a $1.00 chords or the error by the one so then we can we opt in that then we got the best. One asleep with and the combination of the number lurch it's about 200. [00:19:31] 15 on a meter in the dark bottom and the 282300 it's the best for the highest s.p.p. generation so within that design we kept We also using the f. d.t.d. more than in the sensitivity so in this case we can get that. The sensitivity to different are reflected in the exchanges we found that reflect in x. its sensitivity is about $561.00 a meter per reflecting in the x. unit so this this is basically it's a how the transmission we also can predict the transmission with the different sides of their open none of Slate. [00:20:16] It's interesting that a we see the correlation between the picture. From the f.t. d.d. and the shipping imposed of the. S.p.p. generation efficiency which it's correlated to in the near our relationship. That's for sure and so the both of the creation issue in this design would be. Good for the device so then we fabricated a device and measure the transmission spectrum. [00:20:49] Compare there for the actual. Per meter it fits very well so in this case we also measure the actual. Reflect a you next sensitivity it turns out to be. About 500 actually for these the rise Ok We want you to everyone at the top you know our interference or so we pop at the safe pop or go to its sensing mode you may have higher our sensitivity then there have a prompt explosion to. [00:21:30] Analyze So this is a week or 2 they say it's a non optics so we're already seeing these slides so then we wanted to kind of confirm whether this device would be good or for n And I it's the delivery So this suggests there's some combine the data from and they're both the sim and I to go and I says for the s a p r And the definitely results this design for their popped a cap power Sycorax at the top Nonis leadership. [00:22:08] Sensitivity is close to $600.00 and on a major per reflecting index. For this we want you to shoo the protein diffusion into then honestly it we collaborated with the professor in university Akron. His name it's a Dr Smith and then in that case we use in the intro reflection florescence imaging equipment which can only imaging if there's a fluorescent in the anonymously it's close to the bottom of boundary it's like $101.00 on a meter so we see after if like a 40 seconds we've already seen a lot of our protein in the slit I have a video but it's not a walk in the slides I just want to show over here so now you can see some of the flash the protein with the label there for a resonance. [00:23:17] Over there so that about. 40 seconds you already see a lot of protein fusion into them honestly it. In this case then we also use in their fluorescents a correlation spectroscopic to see what's there every time that they're protein can stay in the non-athlete we use a few different types of Nona's lead 100 meter and the $300.00 to $300.00 the open area for there are none allegedly done so the their prime we average in similar time in the normal slayed at the Point 3 few milliseconds we know there are any in the body or and region interactions it's in the would be the normal scale so there would be some fish and for any reaction if for the particle the pair marker can pander to the surface of the. [00:24:25] Function of the surface. So we use in just the fabricated that they say at them and they're cross over them with the non-allergic structural integrity in the single channel microfluidics then we see this is just optical over the lights transmitter to the to the non-allergic area this is a demonstration for. [00:24:48] Prostate a specified antigen detection just as a proof of concept we can see that the transmission we will end shaved with their prostate specific antigen we hear over the then next and then we also wanted to see if it war or that the virus can detect any other like a markers so recently we are chills entally insulin antibody which is a proper marker of a pipe one type Bettys So using this. [00:25:25] As a demonstration we function it's the safe it's used in service and I'm on an air. Their human insulin it's the engine of for the empty insulin anybody basically and and to use and anybody wants it's a high level which is it's a sim a signal over the pipe one pipe at his progress progress so we functional This is their same channel with them and then the one it's the reference and then the other one it's the Nano sleeve area another ledge area so functional it's the human insulin then with the tact we see we monitored their function addition we see a little bit a shift over the. [00:26:12] Of the. Transformation once we have a say face of function addition. Then But once we only did deliver the. Human serum into the. Channel the microflora channel we don't see the. Significand pick shaved in the transmission then we. Tried to use in this device attacked the n.t. easily and about a half hour and unique human human serum we see the. [00:26:54] Picture and long with conservation changes and the see the linear relationship of. Their large very overconcentration which it's a new to the picture effect so this like it's not there months tradition over their normal lurch and that their success with the delivery protein biomarker into the. Cavity for the detection. [00:27:20] And nonspecific impending control we use in. The Baldwin serum abdomen it's a protein it's are a large amount in the product or sample and also use in the Igi for. Another protein we see it's an unspecified into the human insulin so we don't see the significant the picture from a transmission spectra measurements so this is our basic issuing. [00:27:53] For the bailiff of their model lurch really and the with the proper city Cox sides for their both their sampled delivery the end and it's delivery into their normal lurch cavity and their their optical action by by their transmission spectrum measurements. As a summary you know for this study the motivation for the study basically we wanted to win years Tris problematics device towards the integrated the base right now it's in the normal normal structured cheap but we wanted to a few integrated summerlike portable device in the future by integrating optical device for detection and then this. [00:28:41] The device we showing over here it's a versatile property by integrated a Prada monic opto flooding chip then deliver by the delivery in a nice simple as well as the optical device so the design we sure that the composition as of the s.p.r. generation and the definitely simulation group the 2 for such kind of a device and an I says and a design and then with them on streets every kid the device and the Mount St a huge Protein Power market and the syncing unit chip actually we all are recently we also have an ongoing try to modify the surface a use an app tumors and then for more troponin detection but it is some of the preliminary results will lead up a tent. [00:29:37] In the end I would like to give my acknowledgments to you in this therefore we got the n.s.a. or for grants for the past 34 years and also of course the cynic program for the facility support at the Jefferson and. This. This works specifically contributed to from 3 students then. [00:30:05] And their planner and the 10 a member already graduated couple Yes they go right now through a provision in China Associate Professor Taylor actually using this technology just couple years ago a family the company recently got in in s f s a b r s one the project it's ongoing working on that your pony and catching you hope a lot and she just got to her defend our Monday for her appear to be. [00:30:39] In the end I would thank you for your attending and I would take any questions or comments for this presentation thank you again. Thank you Professor very there was actually. Will certainly take any questions people want to help take them into the chair. And I guess Well William I have a couple of questions Lydia understood you when you were talking about the dark side. [00:31:10] You said that the sensitivities are actually fortunately in screed or. What not and what plans the extra Now actually we simulation see this year like at the s.p.p. generation maybe there slope about 4 times however they say it's just this year in the. Simulated this s.p.p. generation but the actual the actual binding. [00:31:41] Ones we have there Ok I would have cried if I hear we don't provide any like a binding in the. Code at the go to save face we just issuing that a s p p. S p poor Poret in generation efficiency seems increased a lot placed on the reflective index changing we kind of give an assumption Ok in the in the normal cavity the whole now Macavity. [00:32:14] The reflecting index changes in the in the whole of cavity but in actual cases actually it's different be pacing it because we only have that the very near the surface so then the practical leeway measure that. Here showing the prod there measured are reflected in x. sensitivity. From the surface of finding that it's about a 600 so it's our it's increased from like a 520 to 596. [00:32:51] Any questions from other folks. And I was going to question So again the theme of this summer series is point of care technologies and you credibility with at the end you know the Nori to make this a true point your device you have to take this you know censorship and integrate it with optics with fluid delivery and all of the actors thing yeah how easy or difficult you think that will be yeah that's a good question I sure the over here kind of. [00:33:28] Just a few years ago. People would develop a such kind of a portable device and also. Us I think packs us as a company in developing their. Portable optical devices for s p r localise it has p r So that's something I think it's it's not a my research focus so it needs collaboration with the companies industry companies all like my students just family the company if a long term goal of course need a lot of investment for. [00:34:09] Either. License others. The kind of detector for for can integrate with our cheap over develop a portable detector but it is a it's a mountain I think it is valid comment. Any any questions anybody else. Ok if not we'll thank you again I don't have a Paul always has recorded applause I don't test that recording but but again we can we can take you virtually And we appreciate your You're presenting today thanks very much thanks to everybody else and and we'll see you in one week for the 4th installment of an offense but by everybody all right.
